Introduction
Probiotic are small single celled living organisms either bacteria or yeast that are ingested to benefit health and help fight disease. A wide variety of different probiotics exists, but most are bacteria of the lactobacillus, Bifidobacterium enterococcus, or exherichin genera or are yeast of the genus saccharomyces (Willey et.al 2011). This practical will aid in determining where the commercial probiotics used by human contain live or viable cultures.
Methods and materials
A probiotic (tablet) was grinded to fine powder and an additional probiotic sachet was used. The antents in of the probiotic sachet, powdered tablet or capsule were used and added to 2x2 ml of milk in sterile Erlenmeyer flasks. A glass rod was used to stir

The pros being: probiotics modify the microbial population of the harmful and harmless microbes, help with digestion and absorption of nutrients. They help in the growth and intestinal cells, improve gastrointestinal issues such as constipation, diarrhoea, and bloating. They can lower hypertension, prevent colon cancer, and they can also reduce periodontal diseases. The cons are: probiotics can cause a boat load of issues if they are not balanced in the system, issues like overgrowth of bacteria or yeast in the gut, which can cause bloating. They can contribute to the indications of rheumatoid arthritis and thrush (yeast infection). It is also known as the klebs –Laffer found in temperature zones but also found in other parts of the world. Diphtheria is an infection caused by this bacteria, is transmitted from one person to another usually through respinary droplet, like from coughing or sneezing.
CONCLUSION
Commercial probiotics contain viable cultures, they are essential to maintain and modify the structures and functions of the intestinal epithelium, and to fight pathogenic bacteria. Diary and fermented products mostly contain probiotics. Probiotics help in reducing some diseases while maintaining others. They are important to life.

Bacteria are living things that have only one cell. Under a microscope, they look like balls, rods, or spirals. Many are helpful. Some bacteria help to digest food, destroy disease-causing cells, and give the body needed vitamins. Bacteria are also used in making healthy foods like yogurt.
Identification of bacteria within Unknown Culture #21 In this experiment, an unknown culture of two different types of bacteria was assigned to each person, a number of tests were performed to isolate and identify these bacterial cells. Based on knowledge from the previous experiments completed in lab, a basic understanding of each type of bacteria was used to create a flow chart that would aid the process of identifying the unknown bacteria within the culture. A gram stain that is performed initially will narrow down the types of tests certain bacteria will and will not respond to. In addition to the gram stain, some of the tests that were used include, a catalase test, an Eosin methylene blue (EMB) agar test, a bile esculin test, and a 6.5% sodium chloride (NaCl) test.
The purpose of this lab report is to employ a myriad of skills, tools and, methods learned throughout this semester to perform the appropriate tests for the identification of the assigned unknown bacteria. The most important tools and techniques used during this identification include aseptic technique, microscopic examination and, the use of selective and differential media. Aseptic technique is an important tool for microbiologists. It is imperative that aseptic technique is maintained throughout the length of any test to avoid any cross-contamination that may lead to inaccurate results.
The author starts by explaining the human microbiome and the research on it. For many years, biologists thought that the human body was able to perform all of its important functions by itself. However, in the past decade, they have found that the human body contains trillions of microorganisms that help with these important processes. In fact, they outnumber human cell 10 to one. At first, microbe research was only focused on harmful pathogens, but recently, biologists have figured out what the most common bacteria in the human body are, and have been trying to figure out the specific functions of theses microorganisms.

They are vitally important at breaking down food and help keep other biota in check. Sometimes antibiotics are taken by patients for issues that are completely unrelated to C. difficile. When the bacteria are given an a chance to grow more than normal, they will pounce on the opportunity, like most other bacteria. When dealing with C. difficile, special precautions must be taken in order to keep it in check and keep ourselves feeling happy and healthy. Humans must understand that we cannot stop all bacteria from doing what they do best, which is grow and adapt to their current environments to their full capability.

In the laboratory, identification of an unknown bacterium is often necessary. In the lab, a random sample consisting of three different bacteria was selected. The sample contained one gram-positive, one gram-negative paracolon, and one gram-negative coliform. The purpose of the experiment is to identify each of the three species that the mixture contained. After receiving an unknown mixture, the sample was streaked for isolation onto TSA, blood agar, and MacConkey plates.
Being able to identify unknown microbes from systematic testing is what makes the field of microbiology so important, especially in infectious disease control. Using the testing procedure laid out by the microbiology field we are able to identify unknown bacteria present in our everyday lives, and along the way learn a lot about their characteristics that separate them from other types of bacteria. Being able to do this is vital in order for us to understand why microbes are present in certain places, how they are able to grow and what restricts their growth, that way they can be combatted if necessary. These techniques for determining unknowns are also important for isolating and testing infectious disease microbes in order to prevent spreading. After a gram stain was done unknown #257 was identified as a gram positive organism because when observed under the microscope the organism appeared purple with cocci in clusters. The organism was also catalase positive which means that it produced enzyme catalase and bubbled when hydrogen peroxide was added to it. Three test were conducted based on the result of the gram staining procedure. Blood agar with a Novobiocin disk was chosen as well as DNase (DNA) and Mannitol Salts (MSA) agar. The Blood agar is a bright red, opaque plate and the streaking or the inoculation technique was a modified streaking for isolation with a heavy quadrant one.

Revision In revising this paper, I took into consideration both deep-level and surface-level revisions. I focused on improving the analysis of my paper by first reading through the essay and thinking over the comments made on both the essay and the grading rubric. From these comments, I realized that I needed to strengthen both my introduction and my conclusion. To achieve this, I reworded the introduction and strengthened my thesis statement to ensure that it was precise and focused on my topic.
