Nigel Courtenay-Luck, Ph.D., CSO of Antisoma, at a recent 9th European Biotech Crossroads meeting, talked about data from Phase 1 of a study on a new type of biomolecule called an aptamer. He said that aptamers are very similar to antibodies; they can both recognize and bind to antigens, they both have low nanomolar binding, and the pKa of both molecules can be modified easily. He said that they are ideal to be used as therapeutics because they can bind well(since they are 3-D) without causing an immunogenic response(since they are small). At this meeting, Dr. Courtenay-Luck showed preclinical and Phase 1 trials for an aptamer called AS1411, an aptamer that recognizes and binds to nucleolin, “a major nuclear phosphoprotein that is found on the surface of many solid tumors” but not really seen on the surface of noncancerous cells. …show more content…
In the Phase 1 study, 17 patients with all sorts of solid tumors were treated for up to 7 days with a dose of 110 mg/kg of AS1411 per day. The results were that there was no “severe toxicity” to the patient seen at levels that would cause them to recover. According to the Corazon y Alma profile, the hospital has a high C-section rate and many of these patients were considered high risk. The hospital has enhanced OB services but access to prenatal care is absent and this has caused a gap in care. There are financial, geographic and social barriers in the community which contribute to the gap.
Studies are being conducted to determine the possibility that the ketogenic diet as a cancer treatment that targets only cancer cells. Lack of glucose essentially keeps cancer cells from dividing as they have no way to gain energy required for the cell cycle. Cells divide to remain small which allows them to function, without being able to divide they will grow until they are forced to commit apoptosis, furthermore, proliferation will decrease when cancer cells lack required energy for division resulting in decreased tumor size. Current cancer treatments, chemotherapy and radiation, are unable to discriminate between healthy and cancerous cells, treatment process is not always affective and can lead to further cancer. Conventional cancer treatment is a multibillion dollar industry.
2.7 Relevance for my personal career development Towards the end of my PhD I dedicated my future research to cancer metastasis, because metastasis is the leading cause of cancer-related deaths. With my future research I want to contribute to a better understanding of this deadly disease. My goal is to improve current mouse models to elucidate novel mechanisms and therapeutic targets of metastasis. The laboratory of Dr. Abate-Shen, which is specialized in developing GEMMs of metastatic prostate and bladder cancer, is an outstanding place for my postdoctoral training and will enable me to learn novel state-of-the-art techniques in the field of metastasis research. My expertise in different mouse models (GEMMs, xenograft) will allow me to spend more time in learning approaches, in which I am not yet an expert (FACS-sorting, computational analysis).
